HuProt utilizes advanced non-contact piezoelectric printing technology, which ensures superior spot morphology and reproducibility compared to traditional contact pin printing methods. Each protein undergoes rigorous purification to select for correctly-folded GST-fusion proteins that maintain their native three-dimensional conformation and biological function. It is designed to deliver remarkably low background noise with exceptional signal-to-noise ratios, enabling reliable high-throughput screening of over 21,000 proteins in a single experiment. This technical precision, combined with the platform's ability to detect both linear and conformational epitopes, provides researchers with robust, reproducible data that accelerates discovery timelines while maintaining the highest standards of analytical accuracy and reliability.
Versatile Applications
Uniquely suited for autoantibody profiling, biomarker discovery, serum profiling, and target identification, enabling researchers to explore protein interactions including both protein-protein and protein-small molecule dynamics
Protein Integrity
Uses properly folded, 3D proteins that retain their native conformation and biological function. This is crucial because many protein interactions depend on proper protein folding
High Reproducibility
Non-contact piezoelectric printing process ensures consistent results with excellent spot morphology and reproducibility across different production batches
High Throughput
Simultaneous analysis of thousands of human proteins in a single experiment, enabling researchers to efficiently screen and profile protein interactions, expressions, and potential biomarkers across various samples
Low Background Noise
Engineered for precision with exceptional signal-to-noise ratios, ensuring reliable high-throughput screening and robust data analysis
CDI Labs’ non-contact piezoelectric ‘inkjet’ process uses next-generation Arrayjet printers and allows for rapid production of high-quality microarray slides time after time. In comparison to older contact pin printing methods, HuProt™ microarrays are made with improved accuracy and reproducibility with excellent spot morphology.
Pooled human plasma was incubated on pairs of HuProt proteome microarrays across two print batches and stained with a fluorescent anti-IgG secondary antibody. Raw median fluorescence intensity data were captured, log2 transformed, and plotted. Linear regression analysis demonstrates reproducibility across the array pair from Lot 1 (R2 = 0.96), Lot 2 ( R2 = 0.92), and the averaged data from both array batches (R2 = 0.96). These results demonstrate the robust reproducibility of HuProt autoantibody seromics data both within and across microarray production lots.
Protein purification prior to microarray manufacturing selects for proteins with correctly-folded GST-fusion proteins. Testing on the HuProt Microarray demonstrates that the proteins are folded in native conformation and retain function (Hu S. poster; Venkataraman A., et al., 2018). When both native and denatured HuProt microarrays were probed with monoclonal antibodies that selectively recognize either linear or folded epitopes of their cognate antigen, the antibodies were found to recognize the appropriate antigen form (Venkataraman A., et al., 2018). Further tests (RNA binding) demonstrated that proteins on HuProt do retain function (Venkataraman A., et al., 2018).
We also offer high-throughput autoantibody signature validation services using focused microarrays. Autoantibody biomarkers targeting up to 300 proteins discovered using HuProt can be arrayed in a format optimized for the size of your biomarker set and throughput requirements. This service offers a fast and affordable solution for biomarker validation. We can synthesize individual proteins at scale for high-throughput screening and other applications.
